SRX23129591: Shotgun metagenomic sequencing of environmental sample
1 ILLUMINA (NextSeq 500) run: 26.8M spots, 8.1G bases, 3.5Gb downloads

Design: We use Picogreen (Invitrogen) to quantitate genomic DNA samples. DNA is sheared using a Covaris and libraries are constructed with the Nugen Ovation Ultralow Library protocol. We aim for an insert size of 400 bp to maximize data. Amplified libraries are visualized on an Agilent DNA1000 chip or Caliper HiSens Bioanalyzer assay, pooled at equimolar concentrations based on these results, and size selected using a Sage PippinPrep 2% cassette. The library pool is quantified using a Kapa Biosystems qPCR library quantification protocol, then sequenced on the Illumina NextSeq in a 1x150 paired-end sequencing run using dedicated read indexing. The samples are demultiplexed with bcl2fastq.

This project targets deep microbial ecosystems of three significant, mostly unexplored (for microbiology) North American geological provinces: the Basin and Range, Black Hills, and Canadian Shield. In addition to their value for the exploration of novel deep habitats, these samples also allow us to address the following questions: (1) Is everything truly everywhere? (2) How does one recognize indigenous deep life? and (3) Is it possible via existing excavations to sample below the lower biosphere limit? These metagenomic datasets complement marker gene sequencing submitted as PRJNA268940. PI Duane Moser
